Recombinant human PTK9 (2-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.

Scientific Background: PTK9 is an actin monomer-binding protein conserved from yeast to mammals which has been localized to cortical G-actin-rich structures that may be regulated by the small GTPase RAC1. PTK9 has tyrosine kinase activity similar to that of recombinant FGFR2, with optimal activity over pH 6.5 to 7.4 and a preference for manganese over magnesium as a divalent cofactor (1). PTK9 plays a central role in regulation of actin dynamics and cell motility in lymphoma cell homeostasis in vivo and represent as a bona fide lymphoma drug target (2).

This gene encodes twinfilin, an actin monomer-binding protein conserved from yeast to mammals. Studies of the mouse counterpart suggest that this protein may be an actin monomer-binding protein, and its localization to cortical G-actin-rich structures may be regulated by the small GTPase RAC1. [provided by RefSeq, Jul 2008]

By capping the barbed ends of filaments, it also regulates motility. Seems to play an important role in clathrin-mediated endocytosis and distribution of endocytic organelles
