Chicken Tumor Necrosis Factor beta ELISA Kit
Principle of the Assay: TNF-beta ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-TNF-beta antibody and an TNF-beta-HRP conjugate. The assay sample and buffer are incubated together with TNF-beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TNF-beta concentration since TNF-beta from samples and TNF-beta-HRP conjugate compete for the anti-TNF-beta antibody binding site. Since the number of sites is limited, as more sites are occupied by TNF-beta from the sample, fewer sites are left to bind TNF-beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TNF-beta concentration in each sample is interpolated from this standard curve.

Lymphotoxin is produced by lymphocytes and cytotoxic for a wide range of tumor cells in vitro and in vivo. Genetic variations in LTA are a cause of susceptibility psoriatic arthritis (PSORAS). PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis).
