Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of general Triglyceride concentrations in serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids.

Principle of the Assay: The ELISA is based on the competitive binding enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Triglyceride, During the reaction, Triglyceride in the sample or standard competes with a fixed amount of biotin-labeled Triglyceride for sites on a pre-coated Monoclonal antibody specific to Triglyceride. Excess conjugate and unbound sample or standard are washed from the plate. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Triglyceride in the samples is then determined by comparing the O.D. of the samples to the standard curve.

LIPC encodes hepatic triglyceride lipase, which is expressed in liver. LIPC has the dual functions of triglyceride hydrolase and ligand/bridging factor for receptor-mediated lipoprotein uptake. [provided by RefSeq, Jul 2008]

LIPC: Hepatic lipase has the capacity to catalyze hydrolysis of phospholipids, mono-, di-, and triglycerides, and acyl-CoA thioesters. It is an important enzyme in HDL metabolism. Hepatic lipase binds heparin. Defects in LIPC are the cause of hepatic lipase deficiency (HL deficiency). A disorder characterized by elevated levels of beta-migrating very low density lipoproteins, and abnormally triglyceride-rich low and high density lipoproteins. Belongs to the AB hydrolase superfamily. Lipase family.
