Applicable Applications for SEC31A sirna
RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of human SEC31A gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

NCBI Description

The protein encoded by this gene is similar to yeast Sec31 protein. Yeast Sec31 protein is known to be a component of the COPII protein complex which is responsible for vesicle budding from endoplasmic reticulum (ER). This protein was found to colocalize with SEC13, one of the other components of COPII , in the subcellular structures corresponding to the vesicle transport function. An immunodepletion experiment confirmed that this protein is required for ER-Golgi transport. Alternative splicing results in multiple transcript variants encoding different isoforms. Uniprot Description

SEC31A: Component of the coat protein complex II (COPII) which promotes the formation of transport vesicles from the endoplasmic reticulum (ER). The coat has two main functions, the physical deformation of the endoplasmic reticulum membrane into vesicles and the selection of cargo molecules. COPII is composed of at least 5 proteins: the SEC23/24 complex, the SEC13/31 complex and SAR1. Interacts with PDCD6 in a calcium-dependent manner. Interacts with KLHL12. Abundantly and ubiquitously expressed. Belongs to the WD repeat SEC31 family. 8 isoforms of the human protein are produced by alternative splicing.

Protein type: Oncoprotein; Vesicle

Chromosomal Location of Human Ortholog: 4q21.22

Cellular Component: Golgi membrane; endoplasmic reticulum membrane; intracellular membrane-bound organelle; COPII vesicle coat; perinuclear region of cytoplasm; endoplasmic reticulum; cytoplasm; ER to Golgi transport vesicle; cytosol

Molecular Function: protein binding; calcium-dependent protein binding

Biological Process: COPII coating of Golgi vesicle; protein transport; ER to Golgi vesicle-mediated transport; antigen processing and presentation of peptide antigen via MHC class I; unfolded protein response, activation of signaling protein activity; cellular protein metabolic process; unfolded protein response; antigen processing and presentation of exogenous peptide antigen via MHC class II; protein amino acid N-linked glycosylation via asparagine; response to calcium ion; post-translational protein modification

Product Notes

The SEC31A sec31a (Catalog #AAA8239261) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The SEC31A siRNA (Human) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SEC31A can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the SEC31A sec31a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SEC31A, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
