Application Notes
1. Run ELISA according to the specific protocol through the conjugate incubation step.
2. Wash the wells three or four times with 1X ELISA Wash Buffer to remove any residual AP-conjugate.
3. Bring SUBP to room temperature; protect from light.
4. Pipette 100 uL SUBP into each well of the plate.
5. Incubate SUBP 10-60 minutes. Monitor the color intensity.
6. Read the plate between 405-420 nm and analyze. Alternatively, stop the reaction by adding 100 uL/well STOPP and read the samples between 405-420 nm within 1 hour.

Store at 2-8 degree C. Protected from light.

Alkaline phosphatase substrates are used in ELISA assays to detect alkaline phosphatase-conjugated molecules via a colorimetric enzymatic reaction. pNPP 1-Component AP Microwell Substrate with Stabilizing Pellets contains para-Nitrophenylphosphate (pNPP). This formulation of pNPP substrate also contains a stabilizer to extend the shelf life of the alkaline phosphatase substrate on your benchtop to 2-3 days.

The alkaline phosphatase substrate reaction may be stopped with the use of a stop solution, Stop Solution for pNPP AP Microwell Substrates.

It is sometimes possible for the material contained within the vial of "pNPP 1-Component AP Microwell Substrate, Substrate" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage.
