The binding activity of RANKL with PPARg.Receptor activator of nuclear factor kappa-B ligand (RANKL), also known as tumor necrosis factor ligand superfamily member 11 (TNFSF11), TNF-related activation-induced cytokine (TRANCE), osteoprotegerin ligand (OPGL), and osteoclast differentiation factor (ODF), is a protein that in humans is encoded by the TNFSF11 gene. RANKL is a member of the tumor necrosis factor (TNF) cytokine family, it binds to RANK on cells of the myeloid lineage and functions as a key factor for osteoclast differentiation and activation. It may also bind to osteoprotegerin, a protein secreted mainly by cells of the osteoblast lineage which is a potent inhibitor of osteoclast formation by preventing binding of RANKL to RANK. Activity Data (Matrix Metalloproteinase 13 (MMP13) is a member of the matrix metalloproteinase (MMP) family. MMP13 has been proposed to participate in aggrecan degradation associated with osteoarthritis and cleavage of type II collagen in osteoarthritic cartilage explants and in tumor progression and metastasis. In addition, it can cleave type I, III, IV, IX, X and XIV collagens and fibronectin. MMP13 is likely to play a crucial role in the modulation of extracellular matrix degradation and cell-matrix interactions. Although gelatin zymography is mainly used for the detection of the MMP2 and MMP9, it also can be used for MMP13 dection. Briefly, various concentrations of MMP13 (1000ng, 500ng, 250ng, 125ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphat- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. Activity Data (PTH (Parathyroid hormone) is a hormone secreted by the parathyroid glands that is important in bone remodeling. As reported, osteoblast-like cell lines, such as ROS 17/2.8, UMR106, SaOS, U2OS, MG63, that exhibit PTHR1, respond with increased proliferation to PTH. Rat PTH shares similarities with human PTH in amino acid sequence with the identity of 71.3%. Thus, a proliferation assay of rat recombinant PTH was conducted using U2OS cells. Briefly, U2OS cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of PTH. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Activity Data (IL7 (Interleukin 7) is a hematopoietic growth factor secreted by stromal cells in the bone marrow and thymus. The interaction between IL17 and the IL7 receptor triggers a cascade of signals important for T-cell development within the thymus and survival within the periphery. It is reported that IL-7 acts on both resting and activated T cells, including Jurkat cells. Thus, a proliferation assay of recombinant human IL7 was conducted using Jurkat cells. Other properties attributed to the cytokine include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. To test the effect of LIF on cell proliferation, TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 including various concentrations of recombinant human LIF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8).
