Activity Data (Protein Kinase R (PKR) is activated by double-stranded RNA (dsRNA), the synthesis of which is caused virally. PKR can also be activated by the protein PACT or by heparin. It plays a key role in the innate immune response to viral infection and is also involved in the regulation of signal transduction, apoptosis, cell proliferation and differentiation. Besides, Cyclin Dependent Kinase 1 (CDK1) has been identified as an interactor of PKR, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PKR and recombinant human CDK1. Briefly, PKR were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CDK1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PKR pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of PKR and CDK1 was shown in Figure 1, and this effect was in a dose dependent manner.)

Western Blot (WB) (Figure 1. Activity Data (Lactoferrin (LF), also known as lactotransferrin (LTF), is a multifunctional protein of the transferrin family. Lactoferrin is one of the components of the immune system of the body; it has antimicrobial activity (bacteriocide, fungicide) and is part of the innate defense, mainly at mucoses. In particular, lactoferrin provides antibacterial activity to human infants. Lactoferrin interacts with DNA and RNA, polysaccharides and heparin, and shows some of its biological functions in complexes with these ligands. LTF is known to activate immune cells to produce several cytokines, such as tumor necrosis factor TNF-a, thus, a stimulation assay was conducted to detect the activity of LTF using human monocytic cell line THP-1. Activity Data (Figure. The binding activity of IFNa with IFNa/bR1.Interferon Alpha (IFNa) belongs to a large subgroup of interferon proteins that help regulate the activity of the immune system. The IFNa proteins are produced by leukocytes. They are mainly involved in innate immune response against viral infection. It is also made synthetically as medication in hairy cell leukemia. Besides, Interferon Alpha/Beta Receptor 1 (IFNa/bR1) has been identified as an interactor of IFNa, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat IFNa and recombinant rat IFNa/bR1. Briefly, IFNa were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IFNa/bR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IFNa pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IFNa and IFNa/bR1 was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Figure. The binding activity of IL33 with IL1RL1.Interleukin 33 is a member of the IL-1 family that potently drives production of T helper-2 (Th2)-associated cytokines. IL33 is a ligand for ST2 (IL1RL1), an IL-1 family receptor that is highly expressed on Th2 cells, mast cells and group 2 innate lymphocytes. IL-33 is expressed by a wide variety of cell types, including fibroblasts, mast cells, dendritic cells, macrophages, osteoblasts, endothelial cells, and epithelial cells. Besides, Interleukin 1 Receptor Like Protein 1 (IL1RL1) has been identified as an interactor of IL33, thus a binding ELISA assay was conducted to detect the interaction of recombinant human IL33 and recombinant human IL1RL1. Briefly, IL33 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL1RL1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL33pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL33and IL1RL1 was shown in Figure 1, and this effect was in a dose dependent manner.)
