(GSK1059615 is a pan-PI3K reversible inhibitor with sub-nanomolar IC50 for PI3Kalpha (0.4 nM) and PI3Kbeta (0.6 nM) and shows low nanomolar activity towards gamma(5 nM),delta(2 nM) and mTOR(12 nM). GSK1059615 inhibits PI3K pathway in cells inducing G1 arrest, although apopto-sis was observed in a subset of cell lines. Breast tumor cells seem to be more sensitive to this compound. In xenograft mice, a complete tumor inhibition and regression were observed together with an increase in plasma insulin levels. [1])
