Scientific Background: SCYL1 is a transcriptional regulator belonging to the SCY1-like family of kinase-like proteins. SCYL1 has a divergent N-terminal kinase domain that is thought to be catalytically inactive, and can bind specific DNA sequences through its C-terminal domain. SCYL1 activates transcription of the telomerase reverse transcriptase and DNA polymerase beta genes. SCYL1 forms multimers following transfection into COS-7 cells (1). A SCYL1 binding protein has been identified that co-localized with SCYL1 in cytoplasm and shows ubiquitous expression (2).

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a transcriptional regulator belonging to the SCY1-like family of kinase-like proteins. The protein has a divergent N-terminal kinase domain that is thought to be catalytically inactive, and can bind specific DNA sequences through its C-terminal domain. It activates transcription of the telomerase reverse transcriptase and DNA polymerase beta genes. The protein has been localized to the nucleus, and also to the cytoplasm and centrosomes during mitosis. Multiple transcript variants encoding different isoforms have been found for this gene. Uniprot Description
Function: Regulates COPI-mediated retrograde traffic. Has no detectable kinase activity in vitro. Ref.8 Ref.11Isoform 6 acts as transcriptional activator. It binds to three different types of GC-rich DNA binding sites (box-A, -B and -C) in the beta-polymerase promoter region. It also binds to the TERT promoter region. Ref.8 Ref.11
Subunit structure: Interacts with GORAB. Interacts with COPA, COPB1 and COPB2
By similarity. Homooligomer. Interacts with AP2B1. Ref.1 Ref.10
Subcellular location: Cytoplasm › cytoskeleton › microtubule organizing center › centrosome. Endoplasmic reticulum-Golgi intermediate compartment. Golgi apparatus › cis-Golgi network. Note: Localized to the Endoplasmic reticulum-Golgi intermediate and cis-Golgi in an ARF1-independent manner. Ref.2 Ref.11Isoform 1: Cytoplasm. Note: Cytoplasmic throughout the cell cycle. Ref.2 Ref.11Isoform 2: Cytoplasm. Note: Cytoplasmic throughout the cell cycle. Ref.2 Ref.11Isoform 3: Cytoplasm. Cytoplasm › cytoskeleton › microtubule organizing center › centrosome. Note: Cytoplasmic during interphase and centrosomal during mitosis, it localizes to the centrosomes in a microtubule-independent manner. Ref.2 Ref.11Isoform 6: Nucleus Ref.2 Ref.11.
Tissue specificity: Ubiquitous. Ref.1
Domain: The protein kinase domain is predicted to be catalytically inactive.
Sequence similarities: Belongs to the protein kinase superfamily.Contains 3 HEAT repeats.Contains 1 protein kinase domain.
Sequence caution: The sequence AAG09726.1 differs from that shown. Reason: Frameshift at positions 289 and 307. The sequence AAG17902.1 differs from that shown. Reason: Frameshift at several positions.

Product Notes
The SCYL1 scyl1 (Catalog #AAA515280) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 1-556 with tag GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant human SCYL1 (1-556) was expressed by baculovirus in Sf9 insect cells. AAA Biotech's SCYL1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the SCYL1 scyl1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SCYL1, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
