Application Notes
Toxoplasma ROP4 antigen is suitable for ELISA and Western blots, excellent antigen for detection of Toxoplasma with minimal specificity problems.

DNA sequence encoding immunodominant fragment of Toxoplasma ROP4 protein was expressed in Escherichia Coli.
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Recombinant TOXGO ROP4 encodes the immunodominant sequences of the Toxoplasma Dense Granule protein.

Recombinant ROP4 contains an N terminal GST purification tag and migrates as an approximately 43 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this protein can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

ROP4, a previously uncharacterized member of the ROP2 family of proteins. ROP4 is secreted from the rhoptries during or shortly after invasion and associates with the PVM, presumably through its predicted transmembrane domain. ROP4 becomes phosphorylated in the infected cell, by either host cell protein kinase (s) or parasite kinase (s) activated by host cell factors.

Function: Thought to play a role in parasitophorous vacuole membrane function during the infection of host organisms. Ref.1

Subcellular location: Secreted. Parasitophorous vacuole membrane. Note: Localized to rhoptries which are secretory organelles at the apical end of the parasite and secreted during host cell invasion when it becomes associated with the parasitophorous vacuole membrane. Ref.1

Post-translational modification: Phosphorylated on multiple serine and threonine residues in parasitic extracts and infected cells but not in extracellular parasites. Ref.1

Sequence similarities: Contains 1 protein kinase domain.

Caution: Lacks the active site residue so is predicted to be catalytically inactive. Ref.1
