Host
Recombinant, Human (E Coli)
Purity/Purification
Highly Purified
98% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified. Endotoxin: 0.1ng/ug (IEU/ug)
Form/Format
Supplied as a lyophilized powder in 10mM sodium citrate, pH 4.5, 150mM sodium chloride. Reconstitute the lyophilized Human Artemin in sterile ddH2O, 0.1% HSA or BSA to 100ug/ml, which can then be further diluted to other aqueous solutions.
Biological Activity
Artemin is fully biologically active when compared to standard. The activity is calculated by it's ability to promote survival and neurite outgrowth and dorsal root ganglion neurons.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm
2. Analysis by RP-HPLC, using a calibrated solution of Artemin as a Reference Standard.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

The protein encoded by this gene is a member of the glial cell line-derived neurotophic factor (GDNF) family of ligands which are a group of ligands within the TGF-beta superfamily of signaling molecules. GDNFs are unique in having neurotrophic properties and have potential use for gene therapy in neurodegenerative disease. Artemin has been shown in culture to support the survival of a number of peripheral neuron populations and at least one population of dopaminergic CNS neurons. Its role in the PNS and CNS is further substantiated by its expression pattern in the proximity of these neurons. This protein is a ligand for the RET receptor and uses GFR-alpha 3 as a coreceptor. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]

Uniprot Description

Function: Ligand for the GFR-alpha-3-RET receptor complex but can also activate the GFR-alpha-1-RET receptor complex. Supports the survival of sensory and sympathetic peripheral neurons in culture and also supports the survival of dopaminergic neurons of the ventral mid-brain. Ref.1 Ref.2

Subunit structure: Homodimer; disulfide-linked

By similarity. Ref.8

Subcellular location: Secreted

Tissue specificity: Ubiquitous. Expressed at high levels in peripheral tissues including prostate, placenta, pancreas, heart, kidney, pituitary gland, lung and testis. Expressed at low levels in the brain. Ref.1 Ref.2

Developmental stage: Expressed during embryogenesis. High level expression seen in fetal kidney and lung while a low level expression seen in the fetal brain. Ref.1

Sequence similarities: Belongs to the TGF-beta family. GDNF subfamily.

Product Notes

The ARTN artn (Catalog #AAA650419) is an Active Protein produced from Recombinant, Human (E Coli) and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "Artemin, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
