Preparation and Storage
Store at 4 degree C if kit is to be used within 1 week. Stable for 6 months (if micro ELISA Plate, Lyophilized Standard and Concentrated Biotinylated Detection Protein stored at-20 degree C. Other components at 2-8 degree C). Stable for 12 months (if the entire kit is stored at-20 degree C).

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- CK- 18/KRT18 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- CK- 18/KRT18 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CK-18/KRT18 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of CK- 18/KRT18 can be calculated.

NCBI Description

KRT18 encodes the type I intermediate filament chain keratin 18. Keratin 18, together with its filament partner keratin 8, are perhaps the most commonly found members of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

Defects are a cause of cryptogenic cirrhosis.
