Intended Uses: This sandwich kit is for the accurate quantitative detection of Rat Troponin C, skeletal muscle (also known as TNNC2) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.
Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Rat TNNC2 antibody. TNNC2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat TNNC2 Antibody is added and binds to TNNC2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated TNNC2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat TNNC2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Troponin (Tn), a key protein complex in the regulation of striated muscle contraction, is composed of 3 subunits. The Tn-I subunit inhibits actomyosin ATPase, the Tn-T subunit binds tropomyosin and Tn-C, while the Tn-C subunit binds calcium and overcomes the inhibitory action of the troponin complex on actin filaments. Troponin is the central regulatory protein of striated muscle contraction. Tn consists of three components: Tn-I which is the inhibitor of actomyosin ATPase, Tn-T which contains the binding site for tropomyosin and Tn-C. The binding of calcium to Tn-C abolishes the inhibitory action of Tn on actin filaments.
