Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-PDGF-AB polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-PDGF-AB polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the PDGF-AB amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of PDGF-AB can be calculated.

Background: Platelet-derived growth factor (PDGF) was one of the first growth factors characterized. and also a potent mitogen for cells of mesenchymal origin, including smooth muscle cells and glial cells. It regulates cell growth and division, In particular, it plays a significant role in blood vessel formation (angiogenesis). It is produced by a plethora of cells, and synthesized, stored, released by platelets upon activation. In chemical terms, PSGF is dimeric glycoprotein composed of two A (-AA) or two B (-BB) chains or a combination of the two (-AB). It is found that initial PDGF-AB interaction with the alpha PDGFR induces conformational changes in the ligand or receptor that facilitates efficient recruitment of beta PDGFR by this PDGF isoform.

NCBI Description

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer (PDGF-BB) or as a heterodimer with the platelet-derived growth factor alpha polypeptide (PDGF-AB), where the dimers are connected by disulfide bonds. Mutations in this gene are associated with meningioma. Reciprocal translocations between chromosomes 22 and 17, at sites where this gene and that for collagen type 1, alpha 1 are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans resulting from unregulated expression of growth factor. Two alternatively spliced transcript variants encoding different isoforms have been identified for this gene. [provided by RefSeq, Feb 2013]

Tissue specificity: Expressed at high levels in the heart, brain (sustantia nigra), placenta and fetal kidney. Expressed at moderate levels in the brain (hippocampus), skeletal muscle, kidney and lung. Ref.20

Involvement in disease: A chromosomal aberration involving PDGFB is found in dermatofibrosarcoma protuberans. Translocation t(17;22)(q22;q13) with PDGFB. Ref.21

Pharmaceutical use: Available under the name Regranex (Ortho-McNeil). Used to promote healing in diabetic neuropathic foot ulcers.
