Intra-Assay: CV<10%
Inter-Assay: CV<12%
Test Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Wide-range Neuregulin 1 (NRG1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Wide-range Neuregulin 1 (NRG1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Wide-range Neuregulin 1 (NRG1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Wide-range Neuregulin 1 (NRG1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

ligand for Erbb3 and Erbb4 receptors; gene produces many different alternative splicing isoforms; involved in neural and organ development [RGD, Feb 2006]

Uniprot Description

Function: Direct ligand for ERBB3 and ERBB4 tyrosine kinase receptors. Concomitantly recruits ERBB1 and ERBB2 coreceptors, resulting in ligand-stimulated tyrosine phosphorylation and activation of the ERBB receptors. The multiple isoforms perform diverse functions such as inducing growth and differentiation of epithelial, glial, neuronal, and skeletal muscle cells; inducing expression of acetylcholine receptor in synaptic vessicles during the formation of the neuromuscular junction; stimulating lobuloalveolar budding and milk production in the mammary gland and inducing differentiation of mammary tumor cells; stimulating Schwann cell proliferation; implication in the development of the myocardium such as trabeculation of the developing heart

By similarity. Ref.6

Subunit structure: The cytoplasmic domain interacts with the LIM domain region of LIMK1. Interacts with ERBB3 and ERBB4. Ref.5

Subcellular location: Pro-neuregulin-1, membrane-bound isoform: Cell membrane; Single-pass type I membrane protein. Note: Does not seem to be active.Neuregulin-1: Secreted.

Tissue specificity: Widely expressed. Most tissues contain isoform alpha2A and isoform alpha2B. Isoform Alpha2 and isoform beta2 are the predominant forms in mesenchymal and non-neuronal organs. Isoform Beta1 is enriched in brain and spinal cord, but not in muscle and heart. Isoform Alpha2C is highly expressed in spinal cord, moderately in lung, brain, ovary, and stomach, in low amounts in the kidney, skin and heart and not detected in the liver, spleen, and placenta.

Domain: The cytoplasmic domain may be involved in the regulation of trafficking and proteolytic processing. Regulation of the proteolytic processing involves initial intracellular domain dimerization.ERBB receptor binding is elicited entirely by the EGF-like domain.

Post-translational modification: Proteolytic cleavage close to the plasma membrane on the external face leads to the release of the soluble growth factor form.N- and O-glycosylated

By similarity. Extensive glycosylation precedes the proteolytic cleavage.

Sequence similarities: Belongs to the neuregulin family.Contains 1 EGF-like domain.Contains 1 Ig-like C2-type (immunoglobulin-like) domain.

Product Notes

The Rat NRG1 nrg1 (Catalog #AAA2709300) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2709300 ELISA Kit recognizes Rat NRG1. AAA Biotech's Neuregulin 1 (NRG1) can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). Researchers should empirically determine the suitability of the NRG1 nrg1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Neuregulin 1 (NRG1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
