Principle of the assay: KMO ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-KMO antibody and an KMO-HRP conjugate. The assay sample and buffer are incubated together with KMO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KMO concentration since KMO from samples and KMO-HRP conjugate compete for the anti-KMO antibody binding site. Since the number of sites is limited, as more sites are occupied by KMO from the sample, fewer sites are left to bind KMO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KMO concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a mitochondrion outer membrane protein that catalyzes the hydroxylation of L-tryptophan metabolite, L-kynurenine, to form L-3-hydroxykynurenine. Studies in yeast identified this gene as a therapeutic target for Huntington disease. [provided by RefSeq, Oct 2011]

KMO: Catalyzes the hydroxylation of L-kynurenine (L-Kyn) to form 3-hydroxy-L-kynurenine (L-3OHKyn). Required for synthesis of quinolinic acid, a neurotoxic NMDA receptor antagonist and potential endogenous inhibitor of NMDA receptor signaling in axonal targeting, synaptogenesis and apoptosis during brain development. Quinolinic acid may also affect NMDA receptor signaling in pancreatic beta cells, osteoblasts, myocardial cells, and the gastrointestinal tract. Belongs to the aromatic-ring hydroxylase family. KMO subfamily. 3 isoforms of the human protein are produced by alternative splicing.
