Test Principle
This assay employs a two-site sandwich ELISA to quantitate G6PD in samples. An antibody specific for G6PD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any G6PD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for G6PD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of G6PD bound in the initial step. The color development is stopped and the intensity of the color is measured.

G6PD: glucose-6-phosphate dehydrogenase. A cytosolic enzyme encoded by a housekeeping X-linked gene whose main function is to produce NADPH, a key electron donor in the defense against oxidizing agents and in reductive biosynthetic reactions. G6PD is remarkable for its genetic diversity. Many variants of G6PD, mostly produced from missense mutations, have been described with wide ranging levels of enzyme activity and associated clinical symptoms. G6PD deficiency may cause neonatal jaundice, acute hemolysis, or severe chronic non-spherocytic hemolytic anemia. Two splice variant isoforms have been described. The long isoform is found in lymphoblasts, granulocytes and sperm.
