Principle of the Assay: The Rat DKK1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat DKK1 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Rat DKK1 coated on a 96-well plate. Standards and samples are pipetted into the wells and DKK1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat DKK1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DKK1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background: Dickkopf-related protein 1 is a protein that in humans is encoded by the DKK1 gene, which was mapped to chromosome 10q11.2. This gene encodes a protein that is a member of the dickkopf family. It is a secreted protein with two cysteine rich regions and is involved in embryonic development through its inhibition of the WNT signaling pathway. Elevated levels of DKK1 in bone marrow, plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma.
