Intended Uses: This sandwich kit is for the accurate quantitative detection of Rat Breast Cancer Susceptibility Protein 1 (also known as BRCA-1) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Rat BRCA-1 antibody. BRCA-1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Rat BRCA-1 Antibody is added and binds to BRCA-1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated BRCA-1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Rat BRCA-1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

The protein encoded by this gene was identified by its ability to bind to the nuclear localization signal of BRCA1 and other proteins. It is a cytoplasmic protein which may regulate nuclear targeting by retaining proteins with a nuclear localization signal in the cytoplasm. [provided by RefSeq, Jul 2008]

Uniprot Description

Negatively regulates MAP kinase activation by limiting the formation of Raf/MEK complexes probably by inactivation of the KSR1 scaffold protein. Also acts as a Ras responsive E3 ubiquitin ligase that, on activation of Ras, is modified by auto-polyubiquitination resulting in the release of inhibition of Raf/MEK complex formation. May also act as a cytoplasmic retention protein with a role in regulating nuclear transport.
