Intended Uses: This AD ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human AD. This ELISA kit for research use only!

Principle of the Assay: AD ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AD antibody and an AD-HRP conjugate. The assay sample and buffer are incubated together with AD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AD concentration since AD from samples and AD-HRP conjugate compete for the anti-AD antibody binding site. Since the number of sites is limited, as more sites are occupied by AD from the sample, fewer sites are left to bind AD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AD concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the ADAM (a disintegrin and metalloproteinase) protein family. ADAM family members are type I transmembrane glycoproteins known to be involved in cell adhesion and proteolytic ectodomain processing of cytokines and adhesion molecules. This protein contains multiple functional domains including a zinc-binding metalloprotease domain, a disintegrin-like domain, as well as a EGF-like domain. Through its disintegrin-like domain, this protein specifically interacts with the integrin beta chain, beta 3. It also interacts with Src family protein-tyrosine kinases in a phosphorylation-dependent manner, suggesting that this protein may function in cell-cell adhesion as well as in cellular signaling. Multiple alternatively spliced transcript variants encoding distinct isoforms have been observed. [provided by RefSeq, Jul 2008]

Suppresses cell motility on or towards fibronectin possibly by driving alpha-v/beta-1 integrin (ITAGV-ITGB1) cell surface expression via ERK1/2 inactivation. Cleaves E-cadherin in response to growth factor deprivation. Plays a role in glomerular cell migration. Plays a role in pathological neovascularization. May play a role in cartilage remodeling. May be proteolytically processed, during sperm epididymal maturation and the acrosome reaction. May play a role in sperm-egg binding through its disintegrin domain. 11 isoforms of the human protein are produced by alternative splicing.
