Principle of the assay: YWHAZ ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-YWHAZ antibody and an YWHAZ-HRP conjugate. The assay sample and buffer are incubated together with YWHAZ-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YWHAZ concentration since YWHAZ from samples and YWHAZ-HRP conjugate compete for the anti-YWHAZ antibody binding site. Since the number of sites is limited, as more sites are occupied by YWHAZ from the sample, fewer sites are left to bind YWHAZ-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YWHAZ concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse, rat and sheep orthologs. The encoded protein interacts with IRS1 protein, suggesting a role in regulating insulin sensitivity. Several transcript variants that differ in the 5' UTR but that encode the same protein have been identified for this gene. [provided by RefSeq, Oct 2008]
