Human Protein Tyrosine Phosphatase Receptor Type N ELISA Kit
Principle of the Assay||PTPRN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PTPRN antibody and an PTPRN-HRP conjugate. The assay sample and buffer are incubated together with PTPRN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PTPRN concentration since PTPRN from samples and PTPRN-HRP conjugate compete for the anti-PTPRN antibody binding site. Since the number of sites is limited, as more sites are occupied by PTPRN from the sample, fewer sites are left to bind PTPRN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PTPRN concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP possesses an extracellular region, a single transmembrane region, and a single catalytic domain, and thus represents a receptor-type PTP. This PTP was found to be an autoantigen that is reactive with insulin-dependent diabetes mellitus (IDDM) patient sera, and thus may be a potential target of autoimmunity in diabetes mellitus.
