Intended Uses: This PRG4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine PRG4. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: PRG4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-PRG4 antibody and an PRG4-HRP conjugate. The assay sample and buffer are incubated together with PRG4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PRG4 concentration since PRG4 from samples and PRG4-HRP conjugate compete for the anti-PRG4 antibody binding site. Since the number of sites is limited, as more sites are occupied by PRG4 from the sample, fewer sites are left to bind PRG4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PRG4 concentration in each sample is interpolated from this standard curve.

Uniprot Description

PRG4: Plays a role in boundary lubrication within articulating joints. Prevents protein deposition onto cartilage from synovial fluid by controlling adhesion-dependent synovial growth and inhibiting the adhesion of synovial cells to the cartilage surface. Defects in PRG4 are the cause of camptodactyly- arthropathy-coxa vara-pericarditis syndrome (CACP); also known as Jacobs syndrome. CACP is an autosomal recessive disorder. Individuals with CACP have normal appearing joints at birth but with advancing age develop joint failure associated with noninflammatory synoviocyte hyperplasia and subintimal fibrosis of the synovial capsule. 6 isoforms of the human protein are produced by alternative splicing.
