This assay has high sensitivity and excellent specificity for detection of SCD. No significant cross-reactivity or interference between SCD and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SCD and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This SCD ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of PorcineSCD. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: SCD ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SCD antibody and an SCD-HRP conjugate. The assay sample and buffer are incubated together with SCD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SCD concentration since SCD from samples and SCD-HRP conjugate compete for the anti-SCD antibody binding site. Since the number of sites is limited, as more sites are occupied by SCD from the sample, fewer sites are left to bind SCD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SCD concentration in each sample is interpolated from this standard curve.

This gene encodes an enzyme involved in fatty acid biosynthesis, primarily the synthesis of oleic acid. Transcripts of approximately 3.9 and 5.2 kb, differing only by alternative polyadenlyation signals, have been detected.
