Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The human WSTF gene is located within the common Williams Syndrome (WS) deletion area at chromosome 7q11.23. Several WSTF gene products have been detected with little difference in length of polypeptides (1-3). Functional motifs identified by sequence-homology searches include a PHD-type zinc finger motif followed by a bromodomain. Both motifs are found in many transcriptional factors, suggesting that WSTF may function as a transcription factor. A Drosophila gene (acf1) was cloned, which encodes two forms of Acf1 proteins with molecular weight 170kD and 185kD, respectively (4). It was demonstrated that Acf1 is structurally related with the human WSTF gene. Acf1 forms a complex with another protein, ISWI, and functions in the ATP-dependent catalysis of chromatin assembly (4).

Uniprot Description

Function: Atypical tyrosine-protein kinase that plays a central role in chromatin remodeling and acts as a transcription regulator. Involved in DNA damage response by phosphorylating 'Tyr-142' of histone H2AX (H2AXY142ph). H2AXY142ph plays a central role in DNA repair and acts as a mark that distinguishes between apoptotic and repair responses to genotoxic stress. Essential component of the WICH complex, a chromatin remodeling complex that mobilizes nucleosomes and reconfigures irregular chromatin to a regular nucleosomal array structure. The WICH complex regulates the transcription of various genes, has a role in RNA polymerase I and RNA polymerase III transcription, mediates the histone H2AX phosphorylation at 'Tyr-142', and is involved in the maintenance of chromatin structures during DNA replication processes. Also involved in vitamin D-coupled transcription regulation via its association with the WINAC complex, a chromatin-remodeling complex recruited by vitamin D receptor (VDR). In the WINAC complex, plays an essential role by targeting the complex to acetylated histones, an essential step for VDR-promoter association
