Western Blot (WB)
(Western blot analysis of extracts from A549, using Wnt3 Antibody. The lane on the left was treated with blocking peptide.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612189 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612189 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Palmitoleoylation is required for efficient binding to frizzled receptors. Depalmitoleoylation leads to Wnt signaling pathway inhibition.
Subcellular Location: Secreted>Extracellular space>Extracellular matrix. Secreted.
Subunit Structure: Forms a soluble 1:1 complex with AFM; this prevents oligomerization and is required for prolonged biological activity (PubMed:26902720). The complex with AFM may represent the physiological form in body fluids (PubMed:26902720).Interacts with PORCN. Interacts with WLS (By similarity).
Similarity: Belongs to the Wnt family.

NCBI Description
The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 98% amino acid identity to mouse Wnt3 protein, and 84% to human WNT3A protein, another WNT gene product. The mouse studies show the requirement of Wnt3 in primary axis formation in the mouse. Studies of the gene expression suggest that this gene may play a key role in some cases of human breast, rectal, lung, and gastric cancer through activation of the WNT-beta-catenin-TCF signaling pathway. This gene is clustered with WNT15, another family member, in the chromosome 17q21 region. [provided by RefSeq, Jul 2008]
Uniprot Description
WNT3: Ligand for members of the frizzled family of seven transmembrane receptors. Wnt-3 and Wnt-3a play distinct roles in cell-cell signaling during morphogenesis of the developing neural tube. Defects in WNT3 are the cause of autosomal recessive tetra-amelia (ARTTRA). Tetra-amelia is a rare human genetic disorder characterized by complete absence of all four limbs and other anomalies such as craniofacial, nervous system, pulmonary, skeletal and urogenital defects. Belongs to the Wnt family.

Product Notes
The WNT3 wnt3 (Catalog #AAA9612189) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Wnt3 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Zebrafish(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(100%), Chicken(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's Wnt3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the WNT3 wnt3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Wnt3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
