Produced in rabbits immunized with E Coli-derived Human UCK1 fragment, and purified by antigen affinity chromatography.
Preparation and Storage
This antibody can be stored at 2-8 degree C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 degree C to -80 degree C. Avoid repeated freeze-thaw cycles.

Immunofluorescence (IF)

(Immunofluorescence staining of UCK1 in U2OS cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human UCK1 polyclonal antibody (dilution ratio 1:200) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus.)

The uridine/cytidine kinase (UCK) family consists of two members, UCK1 and UCK2, and both UCKs are expressed in many tumor cells. UCK1 has an established role in activating 5'-azacytidine (5'-AZA) and its protein level is significantly downregulated in patients resistant to the drug. The KLHL2/USP28/ATM axis mediates resistance of AML cells to 5'-AZA by regulating UCK1 ubiquitination and phosphorylation. UCK2, an enzyme normally expressed in human placenta and testis and highly overexpressed in many neoplasias of blood and solid tissues, catalyzes monophosphorylation of pyrimidine ribonucleosides with efficiency 15-to 20-fold higher than that of ubiquitously expressed isozyme UCK1. Uridine-cytidine kinases (encoded by UCK1, UCKL1, and UCK2) catalyze the phosphorylation of uridine and cytidine to uridine monophosphate (UMP) and cytidine monophosphate (CMP).
