Purified, E. coli-derived, recombinant human TIR domain-containing adaptor inducing IFN-b (aa29-204; Accession #Q8IUC6).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

TRIF (TIR domain-containing adaptor inducing IFN-b; also TICAM1) is a 105-110kD cytoplasmic adaptor molecule that mediates Toll receptor signaling. It is widely expressed and associates with both TLR3 and TLR4. Relative to TLR3, TRIF appears to activate IRF-3, -4, and -7, as well as NFkB and FADD. Its action on FADD is through RIP1, and this induces apoptosis. Human TRIF is 712aa in length and contains three TRAF6 bonding motifs (aa84-91, 248-255 and 299-309), one TIR domain (aa390-460), a Pro-rich region (aa614-678), and an overlapping RHIM domain (aa661-699). The molecule is reported to form a homodimer. There are multiple potential isoform variants. One shows a 23aa substitution for aa31-162 accompanied by a Pro substitution for aa633-660, a second shows a 44aa substitution for aa352-712, and a third shows a 38aa substitution for aa271-712. Over aa29-204, human TRIF shares 62% aa identity with mouse TRIF.

TICAM1: an adaptor protein involved in innate immunity against invading pathogens. Associates with TLR3 and TLR4 (through TICAM2) to mediate NF-kappa-B and interferon-regulatory factor (IRF) activation, and to induce apoptosis. Ligand binding to these receptors results in TICAM1 recruitment through its TIR domain. Distinct protein-interaction motifs allow recruitment of the effector proteins TBK1, TRAF6 and RIPK1, which in turn, lead to the activation of transcription factors IRF3 and IRF7, NF-kappa-B and FADD respectively. Interacts with the TIR domain of TLR3. Interacts with AZI2, TBK1, IRF3 and IRF7. Interacts with TICAM2 in TLR4 recruitment. Interaction with PIAS4 inhibits the TICAM1-induced NF-kappa-B, IRF and IFNB1 activation. Interacts with IKBKB and IKBKE. Interaction with SARM1 blocks TICAM1-dependent transcription factor activation. Interacts with TRAF3. Interacts with TRAFD1.
