Antibody Supershift: An Independent laboratory showed that Anti-STAT5 (A&B) supershifted IL-2 induced complexes in both fresh and preactivated peripheral blood lymphocytes. This antibody also supershifted the complexes induced by IL-2, IL-7 and IL-15, but not those induced by IL-4 and IL-13 in fresh peripheral blood lymphocytes. The specificity of the reaction was confirmed by competition with the immunogenic peptide (Lin, et al., 1995) Experimental conditions must be optimized to achieve the best supershift banding pattern (see Lin, et al., 1993).
Immunohistochemistry: Not tested.

STAT (signal transducers and activators of transcription) proteins can be selectively phosphorylated on tyrosine residues by Jak family kinases and some receptors with tyrosine kinase activity. The STAT proteins are highly conserved in their amino termini but have divergent carboxy termini, which are thought to be essential for their selective activity. STAT5 can be activated by a variety of agents including prolactin, IL-2, IL-3, IL-7, IL-15, growth hormone and erythropoietin. In peripheral blood lymphocytes (PBL), IL-2 rapidly activates STAT5 while phytohemagglutinin activates both STAT3 and STAT5. Evidence indicates that both STAT5A and STAT5B are activated in PBL in vivo. Data show that either Tyr-392 or Tyr-510 of the IL-2Rb is required for IL-2 dependent activation of STAT5. JAK1 and JAK3 may activate each STAT5 isoform by phosphorylation of a single tyrosine residue.

NCBI Description

The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated by, and mediates the responses of many cell ligands, such as IL2, IL3, IL7 GM-CSF, erythropoietin, thrombopoietin, and different growth hormones. Activation of this protein in myeloma and lymphoma associated with a TEL/JAK2 gene fusion is independent of cell stimulus and has been shown to be essential for the tumorigenesis. The mouse counterpart of this gene is found to induce the expression of BCL2L1/BCL-X(L), which suggests the antiapoptotic function of this gene in cells. [provided by RefSeq]
