Immunogen
KLH-conjugated, synthetic peptide corresponding to aa26-39 (C-GEPDPRDALTQQVH) of human Sprouty2, with an N-terminal cysteine added for conjugation purposes. The immunizing sequence is identical in rat and mouse.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Sprouty is a suppressor of EGF and FGF signaling that appears to have a universal inhibitory role on tyrosine kinase signaling. Evidence suggests that it does this by binding adapter proteins and GAP, thus inactivating Ras. Sprouty expression is induced by the same pathways that it down-regulates, suggesting it plays a role in an auto-feedback loop. Sprouty was originally identified as an inhibitor of Drosophila epidermal growth factor (EGF) and fibroblast growth factor (FGF) receptor signaling during tracheal development. Four isoforms of mammalian sprouty are known (Spry1-Spry4). The role of the mammalian analogs has not been clearly elucidated although it is believed that human Sprouty-2 (hSpry2) may be an inhibitor of cellular migration and proliferation. Spry2 and Spry4 show considerable sequence homology between human and mouse at the C-terminus of the protein. Significant sequence divergence occurs at the N-terminus. Spry2 appears to be abundant in brain, lung and heart tissue, with lesser amounts found in kidney and skeletal muscle.

NCBI Description

This gene encodes a protein belonging to the sprouty family. The encoded protein contains a carboxyl-terminal cysteine-rich domain essential for the inhibitory activity on receptor tyrosine kinase signaling proteins and is required for growth factor stimulated translocation of the protein to membrane ruffles. In primary dermal endothelial cells this gene is transiently upregulated in response to fibroblast growth factor two. This protein is indirectly involved in the non-cell autonomous inhibitory effect on fibroblast growth factor two signaling. The protein interacts with Cas-Br-M (murine) ectropic retroviral transforming sequence, and can function as a bimodal regulator of epidermal growth factor receptor/mitogen-activated protein kinase signaling. This protein may play a role in alveoli branching during lung development as shown by a similar mouse protein. [provided by RefSeq]
