Western Blot (WB)
(Western blot analysis of extracts from 293T, using SPRED2 Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612466 at 1/100 staining Rat liver tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)

The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612466 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612466 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Phosphorylated on serine and threonine residues (PubMed:15683364). Phosphorylated on tyrosine. Phosphorylation of Tyr-228 and Tyr-231 are required for ubiquitination (PubMed:17094949).Ubiquitinated; leading to degradation by the proteasome.
Subcellular Location: Cell membrane>Peripheral membrane protein>Cytoplasmic side. Cytoplasmic vesicle>Secretory vesicle membrane>Peripheral membrane protein>Cytoplasmic side. Cytoplasm. Note: Detected in the cytoplasm of the stratum spinosum cells, where it is associated with cytoplasmic vesicles that are supposed to be secretory granules.
Tissue Specificity: Expressed in liver, skin, small intestine, salivary gland and prostate.
Subunit Structure: Homodimer and heterodimer. Able to interact with SPRED1 to form heterodimers (PubMed:15683364). Interacts with Ras (By similarity). May interact with ZDHHC13 (via ANK repeats) and ZDHHC17 (via ANK repeats) (By similarity).

NCBI Description
SPRED2 is a member of the Sprouty (see SPRY1; MIM 602465)/SPRED family of proteins that regulate growth factor-induced activation of the MAP kinase cascade (see MAPK1; MIM 176948) (Nonami et al., 2004 [PubMed 15465815]).[supplied by OMIM, Mar 2008]
Uniprot Description
SPRED2: Tyrosine kinase substrate that inhibits growth-factor- mediated activation of MAP kinase. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Vesicle; Motility/polarity/chemotaxis
Chromosomal Location of Human Ortholog: 2p14
Cellular Component: nucleoplasm; cytoplasm; plasma membrane
Molecular Function: protein serine/threonine kinase inhibitor activity; protein binding; stem cell factor receptor binding; protein kinase binding
Biological Process: multicellular organismal development; positive regulation of DNA damage response, signal transduction by p53 class mediator; inactivation of MAPK activity

Product Notes
The SPRED2 spred2 (Catalog #AAA9612466) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SPRED2 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(88%), Chicken(88%), Xenopus(88%) and may cross-react with other species as described in the data sheet. AAA Biotech's SPRED2 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the SPRED2 spred2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SPRED2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
