Dilution: Western Blot: 3ug/ml. 50ug of NIH3T3 IP cell lysate was resolved via SDS-PAGE, transferred to PVDF and probed with 3ug/ml S1013-33A. Proteins were visualized using an IgG (HRP) anti-rabbit that does not recognize denatured IgG. Detection using a chemiluminescence system.

Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunoprecipitation (IP)

(10ug of MBS619978 was used to immunoprecipitate from 500ug of NIH3T3 whole cell lysate. The ab was collected with Protein A beads and eluted with sample buffer. 50ug of NIH3T3 IP cell lysate was then resolved via SDS-PAGE, transferred to PVDF and probed with 3ug/ml MBS619978. Proteins were visualized using an anti-rabbit (HRP) secondary antibody that does not recognize denatured IgG. ECL detection)

SH3PXD2B is a novel adaptor protein; also known as SH3 and PX domain-containing protein 2B, (Courtneidge, 2005). It is closely related to Tks5 (gene symbol SH3PXD2A) and like Tks5, appears to play a role in the formation and function of podosome/invadopodia based on studies of Src-transformed fibroblasts (Buschman, 2009). Additionally, SH3PXD2B may be involved in the recruitment of MT1-MMP and ECM degradation.

SH3PXD2B is broadly expressed and localizes in the cytoplasm of the podosome.

This gene encodes an adapter protein that is characterized by a PX domain and four Src homology 3 domains. The encoded protein is required for podosome formation and is involved in cell adhesion and migration of numerous cell types. Mutations in this gene are the cause of Frank-ter Haar syndrome. SH3PXD2B: Adapter protein involved in invadopodia and podosome formation and extracellular matrix degradation. Binds matrix metalloproteinases (ADAMs), NADPH oxidases (NOXs) and phosphoinositides. Acts as an organizer protein that allows NOX1- or NOX3-dependent reactive oxygen species (ROS) generation and ROS localization. Plays a role in mitotic clonal expansion during the immediate early stage of adipocyte differentiation. Defects in SH3PXD2B are the cause of Frank-Ter Haar syndrome (FTHS). It is a syndrome characterized by brachycephaly, wide fontanels, prominent forehead, hypertelorism, prominent eyes, macrocornea with or without glaucoma, full cheeks, small chin, bowing of the long bones and flexion deformity of the fingers. Belongs to the SH3PXD2 family.
