Sec1/Munc18-like (SM) proteins, such as SCFD1, are soluble, peripheral membrane proteins that function in distinct intracellular transport steps and interact directly with soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) involved in membrane fusion. Interaction between SCFD1 and the conserved oligomeric Golgi tethering (COG) complex is required for pairing of SNAREs involved in intra-Golgi transport and consequently for Golgi-to-endoplasmic reticulum (ER) retrograde transport. Rat Ra410 contains a consensus sequence associated with vesicle transport-related factors and shares 38% homology with the yeast vesicle transport-related factor Sly1. The SCFD1 gene is associated with amyotrophic lateral sclerosis (ALS).
