Function
Activator of the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors. Upon binding to LGR4-6 (LGR4, LGR5 or LGR6), LGR4-6 associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. Also regulates the canonical Wnt/beta-catenin-dependent pathway and non-canonical Wnt signaling by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway (PubMed: 21909076, PubMed: 21727895, PubMed: 22615920). During embryonic development, plays a crucial role in limb specification, amplifying the Wnt signaling pathway independently of LGR4-6 receptors, possibly by acting as a direct antagonistic ligand to RNF43 and ZNRF3, hence governing the number of limbs an embryo should form (PubMed: 29769720).
Preparation and Storage
Maintain refrigerated at 2-8°C for up to 2 weeks. For long term storage store at -20°C in small aliquots to prevent freeze-thaw cycles.

NCBI Description

R-spondins (RSPOs), such as RSPO2, are secreted proteins that regulate beta-catenin (CTNNB1; MIM 116806) signaling (Kim et al., 2006 [PubMed 16357527]).[supplied by OMIM, Mar 2008]

Uniprot Description

R-spondin-2: Activator of the beta-catenin signaling cascade, leading to TCF-dependent gene activation. Acts both in the canonical Wnt/beta-catenin-dependent pathway and in non-canonical Wnt signaling pathway, probably by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway. Probably also acts as a ligand for frizzled and LRP receptors. Belongs to the R-spondin family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The RSPO2 rspo2 (Catalog #AAA9206616) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 190-218. The RSPO2 Antibody (C-term) reacts with Human (Predicted Reactivity: Mouse) and may cross-react with other species as described in the data sheet. AAA Biotech's RSPO2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB); Immunohistochemistry (IHC-P); Flow Cytometry (FACS/FC); ELISA (EIA). WB~~1:1000 IHC-P~~1:50~100 FC~~1:10~50. Researchers should empirically determine the suitability of the RSPO2 rspo2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RSPO2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
