Western Blot (WB)

(Anti-Rif1 Antibody - Western Blot. Western blot of Affinity Purified anti-Rif1 antibody shows detection of a band ~265 kD corresponding to mouse Rif1 (arrowhead). Specific reactivity with this band is blocked when the antibody is pre-incubated with the immunizing peptide (data not shown). Approximately 25 ug of MEF whole cell lysate was separated by SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1.0 ug/ml for 2 h at room temperature followed by washes and reaction with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX (for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.)

NCBI Description

This gene encodes a protein that shares homology with the yeast teleomere binding protein, Rap1 interacting factor 1. This protein localizes to aberrant telomeres may be involved in DNA repair. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Apr 2010]

Uniprot Description

RIF1: Required for checkpoint mediated arrest of cell cycle progression in response to DNA damage during S-phase (the intra-S- phase checkpoint). This checkpoint requires activation of at least 2 parallel pathways by the ATM kinase: one involves the MRN (MRE11A-RAD50-NBS1) complex, while the second requires CHEK2. RIF1 seems to act independently of both these pathways. Seems to play no role in either the G1/S or G2/M DNA damage checkpoints. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: DNA repair, damage

Chromosomal Location of Human Ortholog: 2q23.3

Cellular Component: chromosome, telomeric region; male pronucleus; cytoplasm; plasma membrane; female pronucleus; spindle; nucleus

Biological Process: stem cell maintenance; cell cycle; response to DNA damage stimulus

Product Notes

The RIF1 rif1 (Catalog #AAA241365) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit Polyclonal to Mouse RIF1 reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's RIF1 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Western Blot (WB), ELISA (EIA). ELISA (1:5000 - 1:40000), IHC-P (5 ug/ml), WB (1:500 - 1:3000) Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary anti.. Researchers should empirically determine the suitability of the RIF1 rif1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RIF1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
