Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Steroid and thyroid hormones and retinoic acid regulate a complex array of gene expression activity via intracellular receptor transcription factors belonging to the ligand dependent nuclear receptor superfamily. Adding to the complexity of function of these transcription factors are associated proteins known as coactivators and corepressors which, as their names suggest, enhance or depress transcription activity of the nuclear receptor with which they associate. Receptor-Interacting Protein 140, also known as RIP140, has been shown to enhance estrogen receptor transcriptional activity through the AF2-AD domain in an agonist dependent manner and has also been implicated to be involved in other steroid receptor pathways, including the retinoid pathway. RIP140 has recently been shown to interact with peroxisome proliferator-activated receptor-alpha (PPARa) and has been proposed to competitively inhibit PPARa interaction with another known nuclear receptor coactivator, SRC-1. However, the specific roles for coactivation and corepression by RIP140 in steroid receptor pathways is currently not well understood.

Interacts with RARA and RXRB homodimers and RARA/RXRB heterodimers in the presence of ligand. Interacts with HDAC1 and HDAC3 via its N-terminal domain. Interacts with NR2C1 (sumoylated form and via the ligand-binding domain); the interaction results in promoting the repressor activity of NR2C1. Interacts with CTBP1, CTBP2, ESR1, HDAC1, HDAC2, HDAC5, HDAC6, NR2C2, NR3C1, NR3C2, YWHAH, JUN and FOS. Found in a complex with both NR3C1 and YWHAH. Interacts with ZNF366.
