Background Info: Detects a ~23kDa protein corresponding to the molecular mass of PUMA on SDS PAGE immunoblots. 16kDa bands may be seen in some instances, possibly corresponding to PUMAbeta.

Scientific Background: Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes. A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 up-regulated modulator of apoptosis) and bbc3 (for Bcl-2 binding component 3) in human and mouse (1-3). PUMA/bbc3 is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-alpha and PUMA-beta (1). PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.

NCBI Description

This gene encodes a member of the BCL-2 family of proteins. This family member belongs to the BH3-only pro-apoptotic subclass. The protein cooperates with direct activator proteins to induce mitochondrial outer membrane permeabilization and apoptosis. It can bind to anti-apoptotic Bcl-2 family members to induce mitochondrial dysfunction and caspase activation. Because of its pro-apoptotic role, this gene is a potential drug target for cancer therapy and for tissue injury. Alternative splicing results in multiple transcript variants. Uniprot Description

CHST9: Catalyzes the transfer of sulfate to position 4 of non- reducing N-acetylgalactosamine (GalNAc) residues in both N-glycans and O-glycans. Participates in biosynthesis of glycoprotein hormones lutropin and thyrotropin, by mediating sulfation of their carbohydrate structures. Has a higher activity toward carbonic anhydrase VI than toward lutropin. Only active against terminal GalNAcbeta1,GalNAcbeta. Isoform 2, but not isoform 1, is active toward chondroitin. Belongs to the sulfotransferase 2 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 2.8.2.-; Membrane protein, integral; Transferase

Chromosomal Location of Human Ortholog: 18q11.2

Cellular Component: Golgi membrane; integral to membrane; extracellular region

Molecular Function: N-acetylgalactosamine 4-O-sulfotransferase activity

Product Notes

The BBC3 bbc3 (Catalog #AAA805746) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The PUMA (CT) Antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's PUMA (CT) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC). 1:1000 (WB), 1:100 (ICC). Researchers should empirically determine the suitability of the BBC3 bbc3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PUMA (CT), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
