Cyclic AMP-dependent protein kinase A (PKA) is an essential enzyme in the signaling pathway of the second messenger cAMP. Through phosphorylation of target proteins, PKA controls many biochemical events in the cell including regulation of metabolism, ion transport, and gene transcription. The PKA holoenzyme is composed of 2 regulatory and 2 catalytic subunits and dissociates from the regulatory subunits upon binding of cAMP.[supplied by OMIM].

NCBI Description

Cyclic AMP-dependent protein kinase A (PKA) is an essential enzyme in the signaling pathway of the second messenger cAMP. Through phosphorylation of target proteins, PKA controls many biochemical events in the cell including regulation of metabolism, ion transport, and gene transcription. The PKA holoenzyme is composed of 2 regulatory and 2 catalytic subunits and dissociates from the regulatory subunits upon binding of cAMP.[supplied by OMIM, Jun 2009]

Uniprot Description

Function: Regulatory subunit of the cAMP-dependent protein kinases involved in cAMP signaling in cells. Ref.9

Subunit structure: The inactive holoenzyme is composed of two regulatory chains and two catalytic chains. Activation by cAMP releases the two active catalytic monomers and the regulatory dimer. Interacts with PRKX; regulates this cAMP-dependent protein kinase. Interacts with C2orf88/smAKAP; this interaction may target PRKAR1B to the plasma membrane. Ref.9 Ref.10

Subcellular location: Cell membrane Ref.10.

Tissue specificity: Four types of regulatory chains are found: I-alpha, I-beta, II-alpha, and II-beta. Their expression varies among tissues and is in some cases constitutive and in others inducible.

Post-translational modification: The pseudophosphorylation site binds to the substrate-binding region of the catalytic chain, resulting in the inhibition of its activity.

Sequence similarities: Belongs to the cAMP-dependent kinase regulatory chain family.Contains 2 cyclic nucleotide-binding domains.

Product Notes

The PRKAR1B prkar1b (Catalog #AAA648017) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The PRKAR1B, NT (PRKAR1B, cAMP-dependent protein kinase type I-beta regulatory subunit) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's PRKAR1B can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in Western Blot, ELISA Dilution: ELISA: 1:1,000 Western Blot: 1:100-500. Researchers should empirically determine the suitability of the PRKAR1B prkar1b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PRKAR1B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
