Interacts (via C-terminal domain) with AKT1; the interaction occurs with the C-terminal cleavage product of PRK2 in apoptotic cells. Interacts (via C-terminus) with PTPN13 (via PDZ 3 domain). Interacts with CDK10. (Microbial infection) Interacts with HCV NS5B (via N-terminal finger domain).
Similarity
The N-terminal regioninterferes with the interaction between AKT1 and the C-terminal regionof PKN2.The C1 domain does not bind the diacylglycerol (DAG).The apoptotic C-terminal cleavage product inhibits EGF-induced kinase activity of AKT1 phosphorylation at 'Thr-308' and 'Ser-473' sites, PDPK1 autophosphorylation and kinases PRKCD and PRKCZ phosphorylations.Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. PKC subfamily.
Subcellular Location
Cytoplasm. Nucleus. Membrane. Cell projection>Lamellipodium. Cytoplasm>Cytoskeleton. Cleavage furrow. Midbody. Cell junction.
Note: Colocalizes with PTPN13 in lamellipodia-like structures, regions of large actin turnover. Accumulates during telophase at the cleavage furrow and concentrates finally around the midbody in cytokinesis. Recruited to nascent cell-cell contacts at the apical surface of cells. In the course of viral infection, colocalizes with HCV NS5B at perinuclear region in the cytoplasm.

Related Product Information for anti-PRK1/2 antibody
PKC-related serine/threonine-protein kinase and Rho/Rac effector protein that participates in specific signal transduction responses in the cell. Plays a role in the regulation of cell cycle progression, actin cytoskeleton assembly, cell migration, cell adhesion, tumor cell invasion and transcription activation signaling processes. Phosphorylates CTTN in hyaluronan-induced astrocytes and hence decreases CTTN ability to associate with filamentous actin. Phosphorylates HDAC5, therefore lead to impair HDAC5 import. Direct RhoA target required for the regulation of the maturation of primordial junctions into apical junction formation in bronchial epithelial cells. Required for G2/M phases of the cell cycle progression and abscission during cytokinesis in a ECT2-dependent manner. Stimulates FYN kinase activity that is required for establishment of skin cell-cell adhesion during keratinocytes differentiation. Regulates epithelial bladder cells speed and direction of movement during cell migration and tumor cell invasion. Inhibits Akt pro-survival-induced kinase activity. Mediates Rho protein-induced transcriptional activation via the c-fos serum response factor (SRF). Involved in the negative regulation of ciliogenesis. (Microbial infection) Phosphorylates HCV NS5B leading to stimulation of HCV RNA replication.

NCBI Description

The protein encoded by this gene belongs to the protein kinase C superfamily. This kinase is activated by Rho family of small G proteins and may mediate the Rho-dependent signaling pathway. This kinase can be activated by phospholipids and by limited proteolysis. The 3-phosphoinositide dependent protein kinase-1 (PDPK1/PDK1) is reported to phosphorylate this kinase, which may mediate insulin signals to the actin cytoskeleton. The proteolytic activation of this kinase by caspase-3 or related proteases during apoptosis suggests its role in signal transduction related to apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been observed. [provided by RefSeq, Jul 2008]

Uniprot Description

PKN1: an AGC kinase of the PKN family. A PKC-related serine/threonine-protein kinase involved in various processes such as regulation of the intermediate filaments of the actin cytoskeleton, cell migration, tumor cell invasion and transcriptional regulation. Regulates the cytoskeletal network by phosphorylating intermediate filament proteins such as VIM and NFH, NFL and NFM, inhibiting their polymerization. Phosphorylates Tau, lowering its ability to bind to microtubules, resulting in disruption of tubulin assembly. Acts as a key coactivator of androgen receptor (ANDR)-dependent transcription, by being recruited to ANDR target genes and specifically mediating phosphorylation of H3T11, a specific tag for epigenetic transcriptional activation that promotes demethylation of histone H3K9me by JMJD2C. Phosphorylates HDAC5, HDAC7 and HDAC9, impairing their import in the nucleus. Activated by lipids, particularly cardiolipin and to a lesser extent by other acidic phospholipids. Activated by CASP3 cleavage during apoptosis. Localization to endosomes is mediated via its interaction with RHOB. Accumulates during telophase at the cleavage furrow and finally concentrates around the midbody in cytokinesis. Three isoforms of the human protein are produced by alternative splicing.

WB: 1:1000-3000 IHC: 1:50-1:200 Peptide ELISA: 1:20,000-1:40,000. Researchers should empirically determine the suitability of the PRK1/2 pkn1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PRK1/2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
