Related Product Information for anti-PRAME antibody
PRAME encodes a 509 amino acid antigen, lacking a signal sequence, and recognized on a human melanoma cell line by a T-lymphocyte clone. A significant level of PRAME mRNA is detected in normal testis as well as in many melanomas, non-small cell lung carcinomas, sarcomas, head and neck tumors and renal carcinomas. PRAME is expressed predominantly in acute leukemias carrying chromosomal abnormalities such as translocation t(8:21), which fuses the AML1 and ETO genes.

NCBI Description

This gene encodes an antigen that is preferentially expressed in human melanomas and that is recognized by cytolytic T lymphocytes. It is not expressed in normal tissues, except testis. The encoded protein acts as a repressor of retinoic acid receptor, and likely confers a growth advantage to cancer cells via this function. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2014]

Uniprot Description

PRAME: Functions as a transcriptional repressor, inhibiting the signaling of retinoic acid through the retinoic acid receptors RARA, RARB and RARG. Prevents retinoic acid-induced cell proliferation arrest, differentiation and apoptosis. Belongs to the PRAME family.

Protein type: Nuclear receptor co-regulator; Cancer Testis Antigen (CTA); Transcription, coactivator/corepressor

Chromosomal Location of Human Ortholog: 22q11.22

Cellular Component: plasma membrane; nucleus

Molecular Function: protein binding; retinoic acid receptor binding

Biological Process: negative regulation of retinoic acid receptor signaling pathway; negative regulation of cell differentiation; transcription, DNA-dependent; apoptosis; regulation of growth; positive regulation of cell proliferation; cell differentiation; negative regulation of transcription, DNA-dependent; negative regulation of apoptosis

Product Notes

The PRAME prame (Catalog #AAA248332) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit Polyclonal (IgG) to Human PRAME reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's PRAME can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Western Blot (WB). IHC-P (7.5 ug/ml), WB (1:500 - 1:3000) Usage: Suggested starting dilutions are as follows: Western blot: 1:500-1:3000. IHC-Paraffin: 7.5 ug/ml. Not yet tested in other applications. Optimal working dilutions should be determined experimentally by the end user. Researchers should empirically determine the suitability of the PRAME prame for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PRAME, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
