Rabbit PPP3CA Polyclonal Antibody | anti-PPP3CA antibody
PPP3CA Antibody
Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol.
IHC: 1:50-1:200
Immunohistochemistry (IHC)
(MBS9606867 at 1/100 staining Mouse liver tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the antibody for 1.5 hours at 22 degree C. An HRP conjugated goat anti-rabbit antibody was used as the secondary.)
Western Blot (WB)
(Western blot analysis of Hela whole cell lysates, using PPP3CA Antibody. The lane on the left is treated with the antigen-specific peptide.)
Function: Calcium-dependent, calmodulin-stimulated protein phosphatase. Many of the substrates contain a PxIxIT motif. This subunit may have a role in the calmodulin activation of calcineurin. Dephosphorylates DNM1L, HSPB1 and SSH1.
Subunit Structure: Composed of two components (A and B), the A component is the catalytic subunit and the B component confers calcium sensitivity (By similarity). Interacts with CRTC2, MYOZ1, MYOZ2 and MYOZ3 (PubMed:11114196, PubMed:11842093). Interacts with DNM1L; the interaction dephosphorylates DNM1L and regulates its translocation to mitochondria (PubMed:18838687). Interacts with CMYA5; this interaction represses calcineurin activity in muscle (By similarity).Interacts with CIB1; the interaction increases upon cardiomyocytes hypertrophy (By similarity). Interacts with CHP1 and CHP2 (By similarity). Interacts (constitutively active form) with SYNPO2 (PubMed:17923693).
Similarity: Belongs to the PPP phosphatase family. PP-2B subfamily.

Uniprot Description
Calcium-dependent, calmodulin-stimulated protein phosphatase which plays an essential role in the transduction of intracellular Ca2+-mediated signals (PubMed:15671020, PubMed:18838687, PubMed:19154138, PubMed:23468591). Many of the substrates contain a PxIxIT motif and/or a LxVP motif (PubMed:17498738, PubMed:17502104, PubMed:23468591, PubMed:27974827, PubMed:22343722). In response to increased Ca2+ levels, dephosphorylates and activates phosphatase SSH1 which results in cofilin dephosphorylation (PubMed:15671020). In response to increased Ca2+ levels following mitochondrial depolarization, dephosphorylates DNM1L inducing DNM1L translocation to the mitochondrion (PubMed:18838687). Dephosphorylates heat shock protein HSPB1 (). Dephosphorylates and activates transcription factor NFATC1 (PubMed:19154138). In response to increased Ca2+ levels, regulates NFAT-mediated transcription probably by dephosphorylating NFAT and promoting its nuclear translocation (PubMed:26248042). Dephosphorylates and inactivates transcription factor ELK1 (PubMed:19154138). Dephosphorylates DARPP32 (PubMed:19154138).

Product Notes
The PPP3CA ppp3ca (Catalog #AAA9606867) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The PPP3CA Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PPP3CA can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), ELISA (EIA). WB: 1:500-1:2000 IHC: 1:50-1:200. Researchers should empirically determine the suitability of the PPP3CA ppp3ca for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PPP3CA, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
