Western Blot (WB) (Figure 1. Western blot analysis of PIAS3 using anti-PIAS3 antibody (MBS1750791). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human Hela cell lysate,Lane 2: human U-87MG cell lysate,Lane 3: human COLO-320 cell lysate,Lane 4: human HepG2 cell lysate,Lane 5: rat brain tissue lysate,Lane 6: mouse NIH3T3 cell lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PIAS3 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for PIAS3 at approximately 68KD. The expected band size for PIAS3 is at 68KD. )

Related Product Information for anti-PIAS3 antibody
Description: E3 SUMO-protein ligase PIAS3 is an enzyme that in humans is encoded by the PIAS3 gene. This gene encodes a member of the PIAS [protein inhibitor of activated STAT (signal transducer and activator of transcription)] family of transcriptional modulators. The protein functions as a SUMO (small ubiquitin-like modifier)-E3 ligase which catalyzes the covalent attachment of a SUMO protein to specific target substrates. It directly binds to several transcription factors and either blocks or enhances their activity. Alternatively spliced transcript variants of this gene have been identified, but the full-length nature of some of these variants has not been determined.
Protein Function: Functions as an E3-type small ubiquitin-like modifier (SUMO) ligase, stabilizing the interaction between UBE2I and the substrate, and as a SUMO-tethering factor. Plays a crucial role as a transcriptional coregulation in various cellular pathways, including the STAT pathway and the steroid hormone signaling pathway. Involved in regulating STAT3 signaling via inhibiting STAT3 DNA-binding and suppressing cell growth. Enhances the sumoylation of MTA1 and may participate in its paralog-selective sumoylation (PubMed: 21965678, PubMed: 9388184). Sumoylates CCAR2 which promotes its interaction with SIRT1 (PubMed: 25406032). Diminishes the sumoylation of ZFHX3 by preventing the colocalization of ZFHX3 with SUMO1 in the nucleus (PubMed: 24651376).

Product Notes

The PIAS3 pias3 (Catalog #AAA1750791) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PIAS3 Picoband antibody reacts with Human, Mouse, Rat No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's PIAS3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 0.1-0.5mug/ml. Researchers should empirically determine the suitability of the PIAS3 pias3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PIAS3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
