This item specifically recognises human PAI-1 (plasminogen activator inhibitor-1), otherwise known as SERPINE1, a 45kDa serine protease inhibitor and member of the serpin family, which acts as the primary inhibitor of tissue plasminogen activator (tPA) and urokinase plasminogen activator (uPA), and hence fibrinolysis.

Increased levels of PAI-1 are associated with diabetes, cardiovascular disease and an increased occurrence of thrombosis in obesity, and can be induced by obesity related factors including insulin, glucose and VLDL (very-low-density lipoprotein).

Reconstitute with 0.1ml distilled water. For long term storage the addition of 0.09% sodium azide is recommended.

Antiserum to human PAI-1 was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

NCBI Description

This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2009]

SERPINE1: a secreted protein that acts as 'bait' for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Belongs to the serpin family. Interacts with VTN. Binds LRP1B; binding is followed by internalization and degradation. Plasma levels of PAI-1 and VCAM-1 together may be useful in predicting post-operative recurrence in patients with colorectal cancer.
