Western Blot (WB) (Figure 1. Western blot analysis of P2RX7 using anti-P2RX7 antibody (MBS1753408).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions.Lane 1: mouse Neuro-2a whole cell lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with rabbit anti-P2RX7 antigen affinity purified polyclonal antibody (Catalog # MBS1753408) at 0. 5 μg/mL overnight at 4 degree C, then washed with TBS-0. 1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # MBS176460) with Tanon 5200 system. A specific band was detected for P2RX7 at approximately 68KD. The expected band size for P2RX7 is at 68KD. )

Preparation and Storage
Store at -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

P2X purinoceptor 7, also called P2X7, is a protein that in humans is encoded by the P2RX7 gene. The product of this gene belongs to the family of purnoceptors for ATP. P2RX7 was mapped to chromosome 12q24 by fluorescence in situ hybridization. It is a receptor for ATP that acts as a ligand-gated ion channel. This gene is responsible for ATP-dependent lysis of macrophages through the formation of membrane pores permeable to large molecules. It could function in both fast synaptic transmission and the ATP-mediated lysis of antigen-presenting cells.

Uniprot Description

P2X7: a receptor for ATP that acts as a ligand-gated ion channel. Responsible for ATP-dependent lysis by macrophages through the formation of membrane pores permeable to large molecules. Activation of this nuclear receptor by ATP in the cytoplasm may be a mechanism by which cellular activity can be coupled to changes in gene expression. Alternatively spliced variants encoding different isoforms have been identified. Could function in both fast synaptic transmission and the ATP-mediated lysis of antigen-presenting cells. Phosphorylation results in its inactivation.
