Western blotting (0.5-4 ug/ml). However, the optimal concentrations should be determined individually. The antibody recognizes mainly ~16 kDa band in human, mouse and rat samples. A ~34 kDa band is also detected to the lesser extent. Reactivity to other species has not been tested.

Background: Noxa is a BH3-only member of the Bcl-2 family. Expression of Noxa gene involves direct activation of its promotor by p53. Increased expression of Noxa protein occurs in normal thymocytes but not in p53-deficient thymocytes. Coimmunoprecipitation data smuggest that Noxa protein may interact with proteins belonging to the Bcl-2 family, such as Bcl-xL and Mcl-1. Blocking the endogenous Noxa induction results in the suppression of apoptosis. These smuggest Noxa may represent a candidate mediator of p53-induced apoptosis.

NOXA: Promotes activation of caspases and apoptosis. Promotes mitochondrial membrane changes and efflux of apoptogenic proteins from the mitochondria. Contributes to p53/TP53-dependent apoptosis after radiation exposure. Promotes proteasomal degradation of MCL1. Competes with BAK1 for binding to MCL1 and can displace BAK1 from its binding site on MCL1. Competes with BIM/BCL2L11 for binding to MCL1 and can displace BIM/BCL2L11 from its binding site on MCL1. Interacts with MCL1, BCL2A1 and BAX. Up-regulated by p53/TP53, phorbol esters, double- stranded RNA, IFNB1/IFN-beta and viruses. Highly expressed in adult T-cell leukemia cell line. Belongs to the PMAIP1 family.
