Specificity
This item recognises Munc-18 phosphorylated at serine 515, a 67 kDa member of the STXBP/UNC-18/SEC1 family. It is neural specific, expressed in the brain, spinal cord and at very high levels in axons. Munc-18 is crucial to calcium-dependent neurotransmitter release, facilitating synaptic vesicle docking and fusion. It inhibits the formation of soluble N-ethylmaleimide-sensitive fusion protein attachment protein receptor (SNARE) complex by binding syntaxin 1 preventing the other components (synaptobrevin and synaptosomal associated protein of 25 kDa (SNAP-25)) from binding. The function of Munc-18 is regulated by the phosphorylation of serine 515 by protein kinase C.

Munc-18 is also involved in the onset of female puberty through its regulation of glutamate release.

Applicable Applications for anti-MUNC-18 antibody
Western Blot (WB)*
Application Notes
Western Blot: This item detects a band/s of approximately 65kDa in rat cortex cell lysates
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Perservative Stabilisers
0.09% Sodium Azide (NaN3)
50% Glycerol
0.01% Bovine Serum Albumin
Immunogen
Phosphopeptide corresponding to amino acid sequence surrounding phosphorylated Ser515 of Munc-18.
Buffer Solution
Antiserum Preparation
Antisera to rat Munc-18 (pSer515) were raised by repeated immunisations of rabbit with highly purified antigen. Purified IgG prepared by affinity chromatography.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.

NCBI Description

inhibits binding of synaptobrevin and SNAP-25 to syntaxin 1A to modulate SNARE complex formation; acts to regulate neurotransmission [RGD, Feb 2006]

Uniprot Description

STXBP1: may participate in the regulation of synaptic vesicle docking and fusion, possibly through interaction with GTP-binding proteins. Essential for neurotransmission and binds syntaxin, a component of the synaptic vesicle fusion machinery probably in a 1:1 ratio. Can interact with syntaxins 1, 2, and 3 but not syntaxin 4. May play a role in determining the specificity of intracellular fusion reactions. Two splice-variant isoforms have been described.

Molecular Function: protein domain specific binding; identical protein binding; protein binding; SNARE binding; syntaxin binding; syntaxin-1 binding; protein N-terminus binding; protein kinase binding

Biological Process: exocytosis; protein stabilization; synaptic vesicle maturation; neurotransmitter secretion; positive regulation of calcium ion-dependent exocytosis; protein transport; platelet degranulation; axon target recognition; neuromuscular synaptic transmission; negative regulation of neuron apoptosis; negative regulation of synaptic transmission, GABAergic; vesicle docking during exocytosis; positive regulation of exocytosis

Product Notes

The MUNC-18 stxbp1 (Catalog #AAA221521) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI MUNC-18 (pSer515) reacts with Bovine (Expected from Sequence), Chicken (Expected from Sequence), Dog (Expected from Sequence), Human (Expected from Sequence), Mouse (Expected from Sequence), Primate (Expected from Sequence), Xenopus (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's MUNC-18 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB)*. Western Blot: This item detects a band/s of approximately 65kDa in rat cortex cell lysates Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background. Researchers should empirically determine the suitability of the MUNC-18 stxbp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MUNC-18, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
