Related Product Information for anti-MAP3K11 antibody
MLK3 (Mixed lineage kinase 3) is a serine/threonine kinase that has an amino-terminal SH3 domain followed by the kinase domain and two leucine zippers, a cdc42/Rac1 binding (CRIB) domain and several other domains/motifs at the carboxy-terminal region. MLK3 functions as a MAPKKK of the SAPK/JNK stress pathway by directly phosphorylating SEK1/MKK4 and MKK7, although it is controversial whether MLK3 is involved in p38 stress pathway activation. MLK3 also functions as an IkB kinase and mediates the activation of the transcriptional factor NF-kB stimulated by CD3/CD28, suggesting a role for MLK3 in immune and inflammatory responses.

NCBI Description

The protein encoded by this gene is a member of the serine/threonine kinase family. This kinase contains a SH3 domain and a leucine zipper-basic motif. This kinase preferentially activates MAPK8/JNK kinase, and functions as a positive regulator of JNK signaling pathway. This kinase can directly phosphorylate, and activates IkappaB kinase alpha and beta, and is found to be involved in the transcription activity of NF-kappaB mediated by Rho family GTPases and CDC42. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Activates the JUN N-terminal pathway. Required for serum-stimulated cell proliferation and for mitogen and cytokine activation of MAPK14 (p38), MAPK3 (ERK) and MAPK8 (JNK1) through phosphorylation and activation of MAP2K4/MKK4 and MAP2K7/MKK7. Plays a role in mitogen-stimulated phosphorylation and activation of BRAF, but does not phosphorylate BRAF directly. Influences microtubule organization during the cell cycle. Ref.1 Ref.5 Ref.9 Ref.10

Catalytic activity: ATP + a protein = ADP + a phosphoprotein. Ref.1

Cofactor: Magnesium

By similarity.

Enzyme regulation: Homodimerization via the leucine zipper domains is required for autophosphorylation and subsequent activation. Ref.6 Ref.7

Subunit structure: Homodimer; undergoes dimerization during activation. Interacts with MAP2K4/MKK4. Interacts with MAP2K7/MKK7

By similarity. Ref.5 Ref.6

Subcellular location: Cytoplasm › cytoskeleton › microtubule organizing center › centrosome. Note: Location is cell cycle dependent. Ref.9

Tissue specificity: Expressed in a wide variety of normal and neoplastic tissues including fetal lung, liver, heart and kidney, and adult lung, liver, heart, kidney, placenta, skeletal muscle, pancreas and brain. Ref.1 Ref.2

Post-translational modification: Autophosphorylation on serine and threonine residues within the activation loop plays a role in enzyme activation. Thr-277 is likely to be the main autophosphorylation site. Phosphorylation of Ser-555 and Ser-556 is induced by CDC42.

Sequence similarities: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase kinase subfamily.Contains 1 protein kinase domain.Contains 1 SH3 domain.

Sequence caution: The sequence BAD92892.1 differs from that shown. Reason: Erroneous initiation.

Product Notes

The MAP3K11 map3k11 (Catalog #AAA646261) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The MLK3 (Mixed Lineage Kinase 3, MLK-3, Mitogen-activated Protein Kinase Kinase Kinase 11, MAP3K11, MEKK11, PTK1, Src-homology 3 Domain-containing Proline-rich Kinase, SPRK) reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's MLK3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Dilution: Western Blot: 0.5-4ug/ml. Researchers should empirically determine the suitability of the MAP3K11 map3k11 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MLK3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
