Testing Data
(Specificity and sensitivity of MBS615382. The antibody reacts specifically with as little as 0.25ng of phosphorylated p42 MAP kinase and does not cross-react with up to 4ug of nonphosphorylated p42 MAP kinase.)
Testing Data
(Confocal images of double immunostaining of rat brain hippocampus (dentate gyrus region), control and 30 minutes of reperfusion following 15 minute transient cerebral ischemia, using MBS615382 (green) and microtubulin associated protein-2 (MAP-2, red). Yellow represents overlay of red and green. (Provided by Dr. Bingren Hu, University of Miami School of Medicine, Florida.))
Testing Data
(Phosphorylated MEK and Erk were assayed in human peripheral blood lymphocytes stimulated with PMA in the presence or absence of the Raf inhibitor BAY 37-9751 or M2865-50 MEK1, 2 Inhibitor. BAY 37-951 blocked PMA-stimulated phosphorylation of both MEK and Erk, consistent with inhibition at the level of Raf, while U0126 blocked phosphorylation of Erk only, consistent with inhibition at the level of MEK. From Chow, S. et al. (2001) Cytometry 46, 72-78.)
Flow Cytometry (FC/FACS)
(Flow cytometric analysis of Jurkat cells, untreated (green) or PMA-treated (blue), using MBS615382 compared to a nonspecific negative control antibody (red).)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the MAP kinase family. MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act in a signaling cascade that regulates various cellular processes such as proliferation, differentiation, and cell cycle progression in response to a variety of extracellular signals. This kinase is activated by upstream kinases, resulting in its translocation to the nucleus where it phosphorylates nuclear targets. Alternatively spliced transcript variants encoding different protein isoforms have been described. [provided by RefSeq, Jul 2008]
Uniprot Description
ERK1: a serine/threonine kinase of the GMGC group that plays a critical role in the regulation of cell growth and differentiation. ERK1 (MAPK3) and ERK2 (MAPK1) play central roles in MAPK cascades and are activated by a wide variety of extracellular signals including growth and neurotrophic factors, cytokines, hormones and neurotransmitters. Depending on the cellular context, MAPK cascades mediate diverse biological functions such as cell growth, adhesion, survival and differentiation through the regulation of transcription, translation, cytoskeletal rearrangements. MAPK cascades also plays a role in initiation and regulation of meiosis, mitosis, and postmitotic functions in differentiated cells by phosphorylating a number of transcription factors. Activation of MAP kinases occurs through phosphorylation of threonine and tyrosine residues at the sequence T*EY* by upstream MAP kinase kinases, MEK1 and -2. Phosphorylation of both the threonine and tyrosine are required for activity. This phosphorylation causes dramatic conformational changes, which enable full activation and interaction of MAPK1/ERK2 with its substrates.

Researchers should empirically determine the suitability of the MAPK3 mapk3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MAP Kinase p44, p42, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
