Cytoplasm > Perinuclear Region. Nucleus. Shuttles between the nucleus and cytoplasm. Found in perinuclear structures together with BCL10.

Immunofluorescene (IF)

(MBS9607520 staining HuvEc by IF/ICC. The sample were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25 degree C. The primary antibody was diluted at 1/200 and incubated with the sample for 1 hour at 37 degree C. An Alexa Fluor 594 conjugated goat anti-rabbit IgG (H+L) Ab, diluted at 1/600, was used as the secondary antibody.)

Description: Mucosa-associated lymphoid tissue translocation gene 1 (MALT1) is a paracaspase that is a critical mediator of T-cell receptor activation of NF-kappaB and may contribute to the progression of MALT lymphomas (1-4). It contains two immunoglobulin-like domains, an amino-terminal death domain and a carboxy-terminal caspase-like domain. Association of MALT1 with Bcl-10 and CARD11/Carma1 leads to activation of IKK and subsequent stimulation of NF-kappaB, resulting in increased proliferation and inhibition of apoptosis (5, 6). A common translocation in MALT B-cell non-Hodgkin lymphomas t(11; 18)(q21; q21) results in the fusion of the amino terminus of API2 (c-IAP2), a member of the inhibitor of apoptosis protein family, to the carboxy terminus of MALT1 (1, 2). The API2-MALT1 fusion protein likely leads to deregulation of NF-kappaB, contributing to increased oncogenic potential (7).
Function: Enhances BCL10-induced activation of NF-kappa-B. Involved in nuclear export of BCL10. Binds to TRAF6, inducing TRAF6 oligomerization and activation of its ligase activity. Has ubiquitin ligase activity. MALT1-dependent BCL10 cleavage plays an important role in T-cell antigen receptor-induced integrin adhesion.
Subunit Structure: Binds through its Ig-like domains to BCL10. Forms oligomers which bind to TRAF6 (PubMed:15125833). Forms a complex with CARD14 and BCL10; resulting in the formation of a CBM (CARD14-BCL10-MALT1) complex (PubMed:27113748). Interacts with BCL10; as part of a CBM (CARD11-BCL10-MALT1) complex involved in NF-kappa-B activation (PubMed:28628108).

NCBI Description

This gene encodes a caspase-like protease that plays a role in BCL10-induced activation of NF-kappaB. The protein is a component of the CARMA1-BCL10-MALT1 (CBM) signalosome that triggers NF-kappaB signaling and lymphoctye activation following antigen-receptor stimulation. Mutations in this gene result in immunodeficiency 12 (IMD12). This gene has been found to be recurrently rearranged in chromosomal translocations with other genes in mucosa-associated lymphoid tissue lymphomas, including a t(11;18)(q21;q21) translocation with the baculoviral IAP repeat-containing protein 3 (also known as apoptosis inhibitor 2) locus [BIRC3(API2)-MALT1], and a t(14;18)(q32;q21) translocation with the immunoglobulin heavy chain locus (IGH-MALT1).
