Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

MAD-1L1 (mitotic arrest deficient 1-like 1) is a member of the spindle-assembly checkpoint (SAC) which prevents anaphase from occurring until all chromosomes are properly aligned at the metaphase plate. Upon checkpoint activation, MAD-1L1 recruits MAD2 to unattached kinetochores, promoting the binding of MAD2 to CDC20, the activator for the anaphase-promoting complex. MAD-1L1 has been implicated to have tumor-suppressing functions, but in small-cell lung cancer cell lines, the MAD-1L1 gene is the most frequent copy number gain, suggesting a more complex role.

NCBI Description

MAD1L1 is a component of the mitotic spindle-assembly checkpoint that prevents the onset of anaphase until all chromosome are properly aligned at the metaphase plate. MAD1L1 functions as a homodimer and interacts with MAD2L1. MAD1L1 may play a role in cell cycle control and tumor suppression. Three transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

MAD1L1: a component of the spindle-assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate. Has a role in the correct positioning of the septum. Required for anchoring MAD2L1 to the nuclear periphery. Expressed weakly at G0/G1 and highly at late S and G2/M phase. Induced by p53. Becomes hyperphosphorylated in late S through M phases or after mitotic spindle damage. Two alternatively spliced isoforms have been described. Defects in MAD1L1 are involved in the development and/or progression of various types of cancer.
